nucleoside reverse transcriptase inhibitor first approved by FDA in 1994 and used for the treatment of adults and children with HIV infection who have undergone prolonged prior AZT therapy. Also called d4T, Zerit.
Indications: Treatment of HIV infection in combination with other agents. Contraindications: Known hypersensitivity, concurrent ZDV use because of pharmacologic antagonism. Dosage: Based on weight: 60 kg -- 40 mg po bid; 60 kg -- 30 mg po bid. Dose adjustment for peripheral neuropathy: 20 mg po bid. Toxicity: Peripheral neuropathy, acute pancreatitis, abnormal liver function tests. Pregnancy category C.
